Dr. Jake Lusis’ research focuses on “systems genetics” approaches to complex disease traits. In these studies, molecular phenotypes, such as transcript levels, are examined globally as a function of genetic variation. These data are then used to model higher order interactions, such as co-expression networks, associated with clinical traits. Experimental manipulations, such as transgenic approaches in mice or siRNA knockdown in tissue culture cells, are used to test hypotheses and to characterize detailed interactions.

A major project in Lusis’ lab is the development of a “Hybrid Mouse Diversity Panel” for systems genetics in the mouse. The panel consists of classic inbred strains of mice as well as recombinant inbred strains. Because the strains are inbred, and therefore permanant, it is possible to carry out many different phenotyping experiments. The panel also offers advantages for mapping, in particular increased resolution as compared to traditional crosses.

Lusis also uses systems genetics to study human cells in culture. For the human studies, his lab has focused on inflammatory responses of macrophages, endothelial cells and smooth muscle cells.
